PROBLEM TO BE SOLVED: To provide a method for adsorbing a polynucleotide having a specific base sequence on a solid-phase surface in a short time and in high selectivity and a method for simply and rapidly judging the base sequence of a polynucleotide to be examined. SOLUTION: The method for selectively adsorbing a polynucleotide having a base sequence part complementary to the base sequence of an oligonucleotide on the solid-phase surface comprises bringing a solution containing the polynucleotide into contact with a solid phase having a surface to which the oligonucleotide is immobilized. The method for selectively adsorbing the polynucleotide comprises cooling a contact surface between the solid phase and the solution from an arbitrary temperature in a range between the melt temperature of a pair composed of the oligonucleotide and the polynucleotide having the base sequence complementary to the oligonucleotide and a temperature 15°C lower than the melt temperature to a temperature at least 10°C lower than the arbitrary temperature at a temperature drop rate of 0.001-0.3°C/c. The presence of the polynucleotide having the specific base sequence part is judged from the adsorbed amount of the polynucleotide adsorbed by the method. COPYRIGHT: (C)2004,JPO
