PURPOSE: A cylinder detection method of an engine is provided to stably control a time for igniting and a time for injecting fuel of the engine by offering stability and reliability in detection of a first cylinder TDC(Top Dead Center). CONSTITUTION: A cylinder detection method of an engine comprises steps of collecting a crankshaft position signal and a camshaft position signal while the engine is in a starting state(S101,S102); determining whether the camshaft position signal detected from a missing tooth of the crankshaft position signal is detected in the order of high, low, and high(S103); determining whether the camshaft position signal is detected in the order of low, high, and low(S104); determining whether the camshaft position is detected in the order of low, low, and high(S105); and re-setting a first cylinder TDC signal detected in the fourth rotation of a crankshaft into a compression stroke by determining whether an exhaust stroke is mistaken for the compression stroke(S106).
