摘要 PURPOSE: A method for screening a compound binding to the active site of a target protein by using specific amino acids labeled protein and 1D NMR technique is provided, thereby easily detecting NMR signal of the active site and decreasing the development cost of proposed medicine. CONSTITUTION: A method for screening a compound binding to the active site of a target protein comprises the steps of: (1) preparing a 15N-labeled target protein and detecting 1D NMR of the target protein; (2) determining the active site signal through a target protein-active site binding compound or target protein-substrate analogue binding test; and (3) carrying out a protein-compound binding test using a compound library to screen a compound showing a change of the active site signal, wherein the target protein is an enzyme or a receiptor; the 15N-labeled target protein is prepared from a recombinant microorganism; and the 15N-labeled amino acids of the target protein include lysine, valine, leucine or isoleucine.
