Detecting specific antibodies (sAb) in a biological sample using the corresponding immobilized antigen (Ag), is new. Detecting specific antibodies (sAb) in a biological sample comprises; (a) culturing host cells; (b) lysing part of these cells; (c) incubating the lysate with the test sample for adsorption of antibodies directed against host cell antigens; (d) incorporating nucleic acid that encodes Ag into a separate portion of the host cells; (e) immobilizing these cells on a carrier; (f) incubating the immobilized cells with the sample of (c) so that specific antibodies (sAb) directed against a recombinant Ag bind; (g) washing the cells and incubation with covalently labeled secondary antibody (Ab2), directed against sAb; and (h) washing the cells and determining bound sAb from the label in Ab2. Independent claims are also included for the following: (1) kit for performing the new method; and (2) solid phase having immobilized on its surface host cells that contain nucleic acid sequences encoding SLA (soluble liver antigen); (3) EBNA-1 (Epstein-Barr nuclear antigen-1) or any of the Treponema pallidum antigens TpN15, N17, N44.5 or N47.
