<P>PROBLEM TO BE SOLVED: To obtain an improved DNA expression system which is useful in production of a protein and a polypeptide, can be used as a recombinant virus or a chimera virus, provides much more merits than those of the prior art and is based on a virus vector. <P>SOLUTION: In order to provide an expression system suitable for producing an effective protein from a cloned DNA in an animal cell, a DNA molecule encoding a protein sequence is inserted into a variant subjected to engineering treatment of cDNA of positive strand RNA virus genome from alpha virus. Then the molecule is used for producing a chimera virus particle for immunization through subsequent RNA transcription and transfection to a tissue culture cell or for producing a recombinant virus for producing a protein. Because of optimization of transfection conditions and properties of virus reproduction, this invention connects simplicity to safety together in terms of operatability, efficiency related to amounts of protein and RNA production and a wide host range. <P>COPYRIGHT: (C)2003,JPO |
