<p>A mammalian cell-infecting virus vector encoding epitope-bound β2m is constructed and thus the epitope-bound β2m is successfully expressed in a large amount in mammalian cells. Namely, it is intended to provide a mammalian cell-infecting virus vector encoding epitope-bound β2 microglobulin (β2m) and utilization thereof. Also, a process for producing an MHC class I/peptide complex with the use of the vector is provided. In particular, a tetramerized MHC class I/peptide complex is useful in detecting epitope-specific CD8-positive T cells. Cells having the above vector transferred thereinto are also provided. Moreover, target cells having epitope-bound β2m produced by the above-described vector added thereto are provided. These cells are recognized by antigen-specific cytotoxic T lymphocytes (CTL). The above vector and the polypeptide obtained via the expression of the vector are useful in immunotherapy for infection and cancer and detection and quantification of antigen-specific CTL.</p> |
