1375001 Enzymatic synthesis of polynucleotides CHOAY SA 8 Nov 1971 [9 Nov 1970] 51775/71 Heading C3 [Also in Division A5] Polynucleotides are obtained enzymatically from the corresponding nucleoside diphosphates using a phosphorylase polynucleotide enzyme (e.g. Azotobacter vinelandii, ATCC 9104), in presence of a magnesium salt, in which the enzyme is added in portions at intervals of time corresponding to the time necesssary for the complete development of the reaction phase which follows the addition of the preceding enzyme fraction. The preferred nucleotides are inosine, cytidine, adenosine, and uridine diphosphates. The resulting polynucleotides may be mixed in equimolar amounts to give Poly I: C and Poly A: U. |
