A method of detection which is based on solution development in which a developer is developed through a test region to a reference region; and a detection reagent for use in the method. The method of detection and the detection reagent are characterized in that the reference region contains a metal compound which is not an alkali metal salt and that a marker which can accumulate in the reference region is contained in the developer reaching the reference region. The marker is preferably fine colored particles. It is preferably one bonded to an antibody or antigen.
