biotechnology. SUBSTANCE: to obtain all-virion vaccine against human cytomegalovirus an AD-169 strain is used. Virus is grown upon a monolayer of diploma cells prelocated into growth medium. Virus cellular releasing is performed at a single cycle of monolayer freezing and thawing. Vaccine purification against detritus is performed by a low-rate centrifuging. As a cellular line one should use diploid cells of human fibroblasts (M-22 line) or renal cellular line of green marmosets VERO-B. Cytomegalovirus is inactivated by formalin at 1:2000 concentration at 37C for 3 d, and at 4°+1° C for 10 d, or by cobalt-60 for 2 h at 19-21 C. Immunogenicity is evaluated by B-cellular index of lymphocytic activity (synthesis of virus-neutralizing antibodies at vaccinating experimental animals). Vaccine is used as a suppository containing, except cytomegalovirus, 20.0 ml (106 tissue cytopathogenic CMV doses) polyoxydonium 0.02 g, hyaluronic acid 1.0 g. Suppositorial base is supplemented with confectionary fat, paraffin and emulsifiers - up to 100.0 g weight.
