PROBLEM TO BE SOLVED: To provide a method of measuring protease activity, using a membrane containing a protease substrate so that expression of the protease activity can be analyzed more precisely than ever. SOLUTION: This method of measuring protease activity comprises (1) a process for bringing a biological sample containing the protease into contact with the membrane, and (2) a for washing the membrane with an aqueous medium and detecting a digestion mark formed on the membrane due to action of the protease, wherein the membrane is formed on the surface of a supporting material and contains at least a kind of pigment selected from an emulsified dispersed pigment and a solidified dispersed pigment together with the protease substrate, and further the membrane is crosslinked and/or is not substantially dissolved in water. Because the method does not require a process for dyeing the membrane, procedures for the measurements are made to be simplified, dyeing of tissues or cells on the membrane is avoidable and therefore the expression of the protease activity can be analyzed more precisely than ever.
