<p>The invention provides a reaction vessel [12] and temperature control system for performing heat-exchanging chemical reactions, such as nucleic acid amplification. The vessel [12] has a body defining a reaction chamber [17], and a loading structure [30] extending from the body for loading a sample into the chamber [17]. The loading structure [30] has a loading reservoir [38], an aspiration port [41], and respective fluid channels [39, 41] connecting the loading reservoir [38] and aspiration port [41] to the chamber [17]. To load the sample into the vessel [12], the sample is first dispensed into the loading reservoir [38] and then drawn into the chamber [17] by application of a vacuum to the aspiration port [38]. The vessel also includes a seal aperture [14] extending over the outer ends of the loading reservoir [38] and aspiration port [41]. A plug [22] is inserted into the aperture [14] after loading the sample into the chamber [17] to simultaneously seal the chamber [17], loading reservoir [38], and aspiration port [41] from the external environment. The temperature of the sample is controlled by opposing thermal plates [50A, 50B] positioned to contact opposite sides of the vessel [12].</p> |
