PROBLEM TO BE SOLVED: To provide a method for observing and determining the localization and dynamic behavior of a protein in an intact cell. SOLUTION: A linearized trap vector, which contains an intron and splice donor sequence, a DNA sequence encoding green fluorescent protein(GFP) from which N-terminal methionine and valine ware deleted, an inner ribosome-binding site, a DNA sequence encoding a selective marker, and a polyadenylated signal sequence, is transfected into a cell, and GFP is expressed as a fused protein with an intrinsic protein. Observation of the green fluorescence allows to observe and determine the localization and dynamic behavior of the intrinsic protein. |
