PROBLEM TO BE SOLVED: To develop both a method for isolating and purifying a nucleic acid such as a plasmid or a genomic DNA from a cell and other raw materials and an apparatus for carrying out the method. SOLUTION: This method for isolating and purifying a nucleic acid comprises (a) removing cell fragments and other particles with a filter layer reducing the pore diameter viewed in the flow direction or the direction to make a sample flow and (b) then treating the resultant eluate with an anion exchanger in a buffer solution at a low ionic strength or a high ionic strength. The apparatus for isolating and purifying the nucleic acid comprises at least one filter layer 12, 20, 21 or 22 arranged on the upstream side of a layer 10 having anion exchange characteristics or a layer 11 capable of binding the nucleic acid in the respective solutions at a high ionic strength and fixed between two means 5 and 6 in the interior of a substantially cylindrical hollow unit 1 when viewed from the direction of an inlet opening 7. |
