A method is disclosed for determining the sequence of a polynucleotide. The sequence is determined by generating the complement of the polynucleotide using the polymerase reaction to extend a suitable primer, and characterising the successive incorporation of bases that generate the complement. The method requires the sequential addition of a composition comprising the different bases A, T, G and C, a minor proportion of which are detectably-labelled. |
