<p>PROBLEM TO BE SOLVED: To make the chromosome specimen of tumor cells such as a myeloma, useful for chromosome examination or the like by culturing tumor cells in the presence of a factor stimulating gp 130 proteins expressed by these cells to propagate the cells and by sampling them. SOLUTION: This chromosome specimen of tumor cells is made by culturing tumor cells such as a myeloma in the presence of a fusion protein of interleukin-6(IL-6) with an IL-6 receptor as a factor stimulating gp 130 proteins expressed by these cells at 37 deg.C and 5% CO2 for 15-24 h. to propagate the cells, treating the cultured cells with colchicine for 1 h., subjecting to hypotonic treatment with 0.075 M KCl for 30 min, fixing with Carnoy fluid, washing the cells twice, dropping a proper quantity of them on a degreased slide and sampling them using an air-drying method. And the examination of the chromosome of tumor cells can be carried out by subjecting the chromosome specimen to G band staining according to a trypsin/Giemsa method to analyze the chromosome after making the chromosome specimen.</p> |
