A synthetic polynucleotide comprising a DNA sequence encoding a non-mammalian protein (such as HIV, HSV, HAV, HBV, HCV, HPV, Plasmodium, Mycobacterium, Borrelia and rotavirus proteins) or fragment is new, in which the sequence comprises codons optimised for expression in a mammalian host. Polynucleotide preferably comprises gene encoding a HIV gag, gag-protease or env gene product and induces anti-HIV neutralising antibodies, HIV-specific T-cell immune responses or protective immune responses when introduced into vertebrate (including in vivo human) tissue. |
