PURPOSE: Improved protective and therapeutic vaccines and improved methods are provided for prophylactically and/or therapeutically inducing immune responses against antigens. CONSTITUTION: A plasmid comprises a nucleotide sequence that encodes a coding sequences operably linked to regulatory elements, wherein the coding sequence encodes an immunogenic target protein linked to or comprising an intracellular targeting sequence.
