PROBLEM TO BE SOLVED: To analyze genome DNAs by treating a genome DNA with the first restriction endonuclease, by subjecting the scission site to the ligation with an adapter, by labeling the adapter, by treating the genome DNA with the second and third restriction endonucleases in sequence and by performing two-dimensional fractionation, followed by detecting spots. SOLUTION: This method comprises the following steps, and enables the mutation of the objective gene, or the like to be detected and the number of spots detected to be controlled, and the analysis of genome DNAs is performed while reducing time, labor and cost needed to screen restriction endonucleases and to set migration conditions, or the like: treating a genome DNA with the first restriction endonuclease; subjecting the scission site to the ligation with the one end of an adapter; labeling the other end of the adapter; treating the DNA fragment with the second restriction endonucleases to perform one- dimensional fractionation; treating the fractionated DNA with the third restriction endonucleases to perform two-dimensional fractionation; detecting spots corresponding to the labeled DNA fragment fractionated by the procedure. |
