METHOD OF PREPARING HUMAN INTERFERON-Ñß 2A

PURPOSE: Provided is a method for manufacturing human alpha interferons 2A in E. coli in a high yield by using an expression vector of arg U gene. CONSTITUTION: A method for manufacturing human alpha interferons 2A in E. coli is characterized by the following steps of: i) implementing PCR with human DNA to obtain gene of human alpha interferons 2A; ii) inserting the gene into restriction enzyme site, Nde I/Bam HI, of plasmid pET-3a to manufacture vector pET-IFN2a; ii) implementing PCR with primers of sequence 3 and 4 described as in the description, to obtain arg U gene and a promoter; iii) inserting the gene and the promoter into restriction enzyme site, BamHI/Eagl, of plasmid pACYC184 to make an expression vector pAC-argU; iv) transforming E. coli BL21 (DE3) with pET-IFN2a and pAC-argU; and v) purifying human alpha interferons 2A from the transformant.
