PROBLEM TO BE SOLVED: To simply and easily obtain sporogenous lactic acid bacteria at a low cost, having high stability in an acidic aqueous solution and useful for various kinds of food and drink or the like by culturing sporogenous lactic acid bacteria in culture medium having a specific acidic pH value in the initial stage of culture and including a specific content of soybean hydrolysate. SOLUTION: The objective sporogenous lactic acid bacteria having high stability in an acidic aqueous solution are simply and easily obtained at a low cost by inoculating sporogenous lactic acid bacterium (Bacillus coagulans) into culture medium having 4.5-6.5 pH value in the initial stage of culture and mixed with 0.01-5.0 wt.% soybean hydrolysate, by the shaking culture at 50 deg.C for 3 days, by centrifuging the obtained culture solution, by washing precipitated spores with 50 mM phosphate buffer showing pH 7, by heat treatment at 80 deg.C for 30 min to make nurse cells become extinct, by suspending spores centrifuged again into 10% glycerin solution, by inoculating the suspension into medium solution showing pH 3. 5 and culturing the spores at 50 deg.C for 14 days. |
