<p>PROBLEM TO BE SOLVED: To produce Schwann cells in a large amount to provide the Schwann cells necessary for the extension of nerves. SOLUTION: This method for purely culturing Schwann cells comprises subjecting the peripheral nerves of an adult mammal to a Wallerian degeneration, collecting the nerves, dispersing the cells with collagenase, culturing the dispersed cells in a culture medium containing a serum by the use of a fibronectin-coated culture dish, confirming the adhesion of fibroblasts, collecting non-adhered Schwann cells, and subsequently culturing the collected Schwann cells in a laminin-coated culture dish. When the contamination of fibroblasts are recognized in the culture medium in the culture dish, the Schwann cells are selectively collected with a buffer solution containing a calcium chelating agent, and subsequently transferred to a new laminin-coated culture dish to enhance the purity of Schwann cells.</p> |
