The present invention includes peptides that bind to and inhibit the function of the integrin receptor, alpha V beta 3. It also includes peptides that bind the integrin receptor alpha V beta 5. The inventive peptides are between nine to about thirty amino acids in length. The peptides include an amino acid sequence comprising the binding domain Thr-Cys-X1-X2-Arg-Ala-Asp-Cys-X3 and an amino acid sequence comprising the binding domain Arg-Cys-Gly-Gly-Asp-Ser-X4-Cys-Tyr. These non-RGD peptides display surprisingly potent antagnonist activity despite the lack of the consensus binding sequence Arg-Gly-Asp, and present opportunities for selective targeting to the alpha V beta 3 receptor. Pharmaceutical compositions and methods of use are also disclosed. The therapeutic uses for the inventive peptides include treating diseases involving alpha V beta 3 receptors such as cancer, osteoporosis, restenosis, and angiogenic-based diseases.
