A method of diagnosis comprises determining genotype of a TGF- beta 1 promoter-specific polmorphisms that have been identified are at positions -800bp and -509bp. The genotype determined is then used to assess predisposition or susceptibility to a number of diseases: osteoporosis, atherosclerosis, cancer and immune disorders.
