Determination of effect of reactive oxygen intermediates on target molecule using cell-free system

Method for determining the effect of reactive oxygen intermediates on a target molecule comprises a detection system comprising the target molecule and soluble components of a eukaryotic cell lysate, introducing the reactive oxygen intermediates into the detection system, and determining the effect of the reactive oxygen intermediates on the target molecule. Independent claims are also included for the following: (1) use of nucleotide analogs containing gamma thio groups, especially ATP- psi S or GTP- psi S, to inhibit activation of iron regulatory protein IRP-1 by reactive oxygen intermediates; and (2) use of phosphorylation and/or dephosphorylation effectors for modulating the effects of reactive oxygen intermediates on target molecules.
