PROBLEM TO BE SOLVED: To detect the high virus level such as a parvovirus B19, etc. in a blood plasma or a blood serum by limiting the sensitivity of a polymerase chain reaction(PCR) using suboptimal nucleic acid extraction, amplification or detection conditions. SOLUTION: The sensitivity of a polymerase chain reaction(PCR) is limited by using suboptimal nucleic acid extraction, amplification or detection conditions to thereby detect the high virus level, especially a parvovirus B19, etc. in a blood plasma and/or a blood serum using the PCR. Furthermore, the suboptimal amplification conditions for the PCR for detecting a parvorirus DNA are established by annealing of a primer to a DNA single strand so that only the parvovirus DNA in a sample at >=10<6> to 10<7> genome equiv./ml content of the DNA can be detected. Thereby, the amplification is carried out at about 52 deg.C in <30 cycles to remarkably limit the sensitivity. As a result, the parvovirus DNA is detected. |
