PROBLEM TO BE SOLVED: To provide the subject new DNA composed of a DNA encoding a protein having a specific amino acid sequence and heat-stable glycerol kinase activity and usable e.g. for the production of the above enzyme useful for the determination of neutral fat in serum, etc. SOLUTION: This new DNA encodes a protein composed of an amino acid sequence expressed by the formula or a protein composed of an amino acid sequence of the formula wherein one or several amino acids are depleted, substituted or added and having a heat-stable glycerol kinase activity comparable to that of the protein of the formula. The DNA is useful e.g. for the production of an enzyme for the determination of neutral fat (triglyceride) level in the serum as an important index for the diagnosis of hyperlipidemia, arteriosclerosis, etc. The DNA can be produced by the Southern analysis of a genom DNA of ultra-high thermophilic archaebacteria Pyrococcus sp.KOD1 strain by using a DNA fragment having amplified glycerol kinase as a probe. |
