PROBLEM TO BE SOLVED: To restrict a running cost by a method wherein a reaction between a DNA sample and a reagent is caused in a fine capillary tube and subsequently, a separation/ detection process is carried out in the same capillary to reduce the amount of the sample and the reagent demanded by the reaction process in the DNA base sequencing. SOLUTION: A capillary 1 for separation is inserted into a sample both 5, a voltage is applied by a high voltage source 9 between a cathode electrode 7 and an anode electric field bath 8 and a sample is injected into the capillary 1. After the capillary is allowed to stand to diffuse, the reagent is caused to react with the sample and a temperature for polymerizing reaction is applied by a temperature control mechanism 10 to the injection side of the capillary 1. The injection side of the capillary 1 and the cathode electrode 7 are placed into a cathode electrode bath 6 and when a migration voltage is applied between the cathode electrode bath 6 and the anode electric field bath 8 by a high voltage power source 9, the sample is migrated in the capillary. A detecting section 2 is irradiated with a laser light 3 and the fluorescence emitted is read by a detector 11 to measure the position of bases in the sample. The results are analysed by a data processing unit 12, thereby determining a base sequence of DNA. |
