摘要 PROBLEM TO BE SOLVED: To obtain the subject new vector containing a DNA having a foreign expression-regulating region and containing the structural gene of a physiologically active substance at both the ends, useful for substituting an expression-regulating region by the foreign expression-regulating region, and giving a human liver cell capable of stably and higher producing the physiologically active substance. SOLUTION: In this new homologous recombination targeting vector for substituting an expression-regulating region for a physiologically active substance by a foreign expression-regulating region by a homologous recombination method, a DNA containing the foreign expression-regulating region, and a DNA having homology for a part of the gene sequence of the 5'-upstream region of a structural gene for the physiologically active substance or the 5'-upstream region containing a part of the 5'-end side of the structural gene are arranged. The targeting vector is useful for producing a human liver cell capable of stably and highly producing the physiologically active substance. The vector is obtained by combining glutathione-S-transferase enhancer with another promotor and subsequently binding the homologous region of a desired physiologically active substance to the 5' and 3'-sides of the combination product.
