Preparation of a non-radioactively labelled nucleoside-3'-phosphate (I), from a nucleoside-3'-phosphate (II) containing a nucleo-base, a pentose and a phosphate group, comprises: (a) coupling (II) with a phosphoramidite (III) containing a non-radioactive label, and (b) oxidising the product. Also claimed is a method for determining modifications of nucleic acids (NA's) by agents (A), comprising: (a) incubating NA with (A); (b) degrading the NA to (II); (c) non-radioactively labelling (II) as described above, and (d) separating and detecting the obtained (I).
