PROBLEM TO BE SOLVED: To obtain a new subject enzyme having a specific amino acid sequence or an amino acid sequence in which a mutation is present in a part, derived from a plant, being a target enzyme of a light requiring type herbicide and useful for creating a plant having a gene regnant to the light requiring type herbicide. SOLUTION: This new protoporphyrinogen oxidase comprises a peptide having an amino acid sequence expressed by the formula or a mutated peptide having the amino acid sequence, in which defects, additions or substitutions, etc., of one or plural amino acids are observed, and enzyme activity essentially the same as that of protorphyrinogen oxidase. The oxidase and its derivative are each a target enzyme of a light requiring herbicide. A plant resistant to the light requiring type herbicide is created by manifesting a large amount of the enzyme in the plant, The enzyme is obtained by isolating mRNA from Arabis thaliana, preparing a cDNA library by using the mRNA and screening to obtain a gene, which is manifested in host cells. |
