PURPOSE: To obtain the subject primer used-for polymerase chain reaction for sex discrimination of a bovine embryo by selecting a base sequence having a specific length and a specified interregional distance without causing a great partial bias to distributions of A, T, G and C in each region from a male- specific base sequence. CONSTITUTION: Two regions satisfying conditions of (1) 15-30 bases length of each region, (2) 40-60% ratio of (G+C) in each region, (3) without causing a great partial bias to distributions of A, T, G and C in each region, (4) 100-1000 bases each interregional distance and (5) the absence of a complementary sequence part in each region itself or between two regions are selected to produce a single-stranded DNA having the same base sequence as that of the region or complementary to the region. Furthermore, a single-stranded DNA modified so as not to lose the binding specificity for the male-specific sequence, as necessary, is produced to afford a primer used for polymerase chain reaction(PCR) for the sex discrimination of a bovine embryo. |
