PURPOSE:To obtain a method for sorting out a homologous recombinant cell, applicable even to a gene unexpressed with an embryonic undifferentiated (ES) cell and having a high sorting out efficiency. CONSTITUTION:The objective method for sorting out a homologous recombinat cell is characterized by using a genome DNA comprising a neo gene having a PGK promoter on the upstream side thereof and an MVM pause signal and a blue script DNA inserted into a region between the neo gene and an MC1 promoter of a diphtherial toxin A fragment gene in a method for homologously recombining the genome DNA in which the neo gene and the diphtherial toxin A fragment gene having the MC1 promoter integrated therein in an embryonic undifferentiated (ES) cell and sorting out a cell containing a genome DNA having a deficient or modified gene therein with G418 and diphtherial toxin A.
