<p>PURPOSE:To readily obtain a cell derived callus to which adventitious genes are transduced by tranducing adventitious genes to guard cells in epidermal tissue of plant by micro injection method, culturing the cells and selecting only green part containing chloroplast. CONSTITUTION:A leaf is immersed in a hypertonic solution, subjected to plasmolysis to be readily micro injected, adventitious genes (a) are transduced to guard cells 3 of epidermal tissue 1 of leaf by micro injection method using a micro pipette below a microscope attached to a manipulator, the epidermal tissue is released from other tissue by tweezers, placed on an agar medium 7 in a flask 6 and subjected to callus culture 8. Since the epidermal cell 2 in the epidermal tissue 1 does not contain chloroplast, green part containing chloroplast is selected to give guard cell derived callus 8a to which adventitious genus are transduced.</p> |
