摘要 PROBLEM TO BE SOLVED: To obtain a tumor necrosis factor(TNF)-suppressing protein having a bonding-suppressive ability of TNF to a receptor and a cytotoxicity- suppressive ability of TNF and usable against a harmful effect of TNF. SOLUTION: This TNF-suppressing protein having an amino acid sequence of the formula (X is an amino acid residue) at an N-end by culturing a transforming obtained by transforming a host cell with a manifestation vector containing a DNA molecule coding a TNF-suppressing protein obtained from a concentrate of a human urine. A TNF-suppressing protein has following properties other than the above: when a crude controlled material of urine is subjected to a chromatography using Vitrogel ACA 44gel permeation column, a main peak of the TNF-suppressing protein is eluted out just before a main portion, having about 40-80kDa apparent molecular weight and pH6-8 isoelectric point of an active protein in analyzing the crude controlled material of urine. The TNF-suppressing protein is useful as a remedy component for an autoimmune disease, etc., such as septic shock, cachexia, graft host reaction and rheumatoid arthritis.
