The present invention relates to DNA structure-specific proteins. Specifically it relates to the nucleotide sequence encoding DNA structure-specific nucleases which have been modified so as to change a codon for at least one amino acid such that the nuclease encoded by the nucleotide sequence is less toxic to a host cell than said wild-type enzyme, and/or said nuclease does not have the catalytic activity of the wild-type enzyme but which retains its structure-selective DNA binding properties of the wild-type enzyme.
