PURPOSE: To enable exact amplification of a nucleic acid in a shortened time by allowing a primer and 4 kinds of dNTP to react with the target nucleic acid in a buffer solution containing a specific heat-resistant DNA polymerase to anneal the primers to the target nucleic acid. CONSTITUTION: A target nucleic acid is modified, when necessary, and dissociated into a single strand and this single strand nucleic acid is allowed to react in a buffer solution containing the A-terminal and N-terminal primers having the base sequence complementary to the target nucleic acid, 4 kinds of dNTP and a heat-resistant DNA polymerase having at least 30 bases/second rate of DNA synthesis and 3'-5'-exonuclease activity to anneal the primer onto the single strand nucleic acid and extend the primers, then the extended primer is dissolved to single strand. These operations are repeated to efficiently amplify the target nucleic acid in high exactness in a shortened time. |
