<p>An affinity ligand (6) such as an antibody or enzyme, or a cell receptor complement is covently bonded to one end of a leash (4) formed of a difunctional spacer and/or bifunctional linking agent that has another end covalently bonded to interior pore surfaces (3) of a high flux semipermeable hydrogel membrane (1) to produce an affinity support for separation of biological molecules such as proteins, cells or cell fragments from solution. Pores of the membrane have an exclusion limit (molecular weight cut-off) selected to substantially restrict immobilized protein or other ligand to the surface of the membrane for maximization of available ligand binding capacity. The exclusion limit is also selected to permit the leash used for bonding of a ligand to the membrane to penetrate into pores and bond to interior pore surfaces of the membrane.</p> |
