PURPOSE: To obtain the subject enzyme having a specific amino acid sequence, producible by gene manipulation technique, active to convert big endothelin into endothelin and useful for the search and evaluation of human endothelin converting enzyme inhibitor, the diagnosis of myocardial infarction, etc. CONSTITUTION: This new human endothelin converting enzyme(ECE) protein is producible by gene manipulation technique, free from other human-originated protein, contains amino acid sequence from the 1st to 758th amino acids in an amino acid sequence containing the amino acid sequence of the formula and is active to convert big endothelin into endothelin. The other new ECE protein has a structure of the above protein with the exception of the depletion, insertion or substitution of one or more amino acid residues at one or more sites of the protein. The protein can be used for the search and evaluation of an ECE inhibitor, the production of a transformant expressing human ECE or an ECE-specific antibody using a part of the transformant as an immunogen and the diagnosis of diseases such as myocardial infarction. |
