PURPOSE: To provide a method for efficiently producing a human thrombomodulin having promoting actions on the activation of protein C with thrombin useful as a therapeutic agent for diseases accompanied by blood coagulation disorder, e.g. cardiac infarction, thrombosis, embolism, peripheral vascular occlusion, obstructive arteriosclerosis, disseminated intravascular coagulation(DIC), angina pectoris, transient cerebral ischemic attack and toxemia of pregnancy. CONSTITUTION: This method for producing a human thrombomodulin is to respectively transduce a DNA sequence I capable of coding a protein, containing all or a part of a human thrombomodulin gene and having thrombomodulin activities, and a DNA sequence II, capable of coding a dihydroflate reductase (DHFR) providing methothexate(MTX) tolerance into an animal cell, cotransform the animal cell, then select the cotransformed animal cell under conditions for progressively increasing the MTX and culture the resultant animal cell. |
