PURPOSE: To obtain the subject new gene having a specific amino acid sequence, coding phosphoenolpyruvic carboxylase, useful as a reagent for measuring a carbon dioxide gas as an enzyme for clinical examination agent, capable of producing the enzyme having excellent stability. CONSTITUTION: This new gene has an amino acid sequence containing an amino acid sequence of the formula, codes phosphoenolpyruvic carboxylase, is useful as a reagent for measuring a carbon dioxide gas as an enzyme for clinical examination and is capable of producing the enzyme having excellent stability. The gene is obtained by separating chromosomal DNA from Acetobacter hansenii IFO14,820 by a conventional method, treating it with a restriction enzyme, bonding a formed fragment to a vector, transducing the vector to Escherichia coli, screening the prepared transformant with an antibody, selecting a clone containing phosphoenolpyruvic carboxylase gene, recovering the DNA and treating it with a restriction enzyme. |
