PURPOSE:To obtain the enzyme useful as a reagent for clinical laboratory, capable of determining ammonia approximately in neutrality and quantitatively measuring ammonia, NAD, etc. CONSTITUTION:This glutamic dehydrogenase is obtained by culturing Bacillus sp. KSM-635 strain (FERM BP-1,485) and has the following physical and chemical properties. (1) Catalyzing reaction forming ammonia, 2-ketoglutaric acid and NADPH from glutamic acid and NADP and its reverse reaction but not acting on NADP and DADPH. (2) The optimum pH is pH about 7.5 (NADPH substrate) and pH about 8.5 (NADP substrate). (3) The optimum temperature is 60-70 deg.C. (4) Km value is 0.01-0.09mM. (5) Molecular weight is 310,000+ or -10,000 (gel filtration method). (6) Isoelectric point is about 4.9. |
