<p>PURPOSE:To obtain a large quantity of seedlings from a tissue of Culculigo plant by the plant tissue culture method, rapidly and without being influenced by the climate, the soil or the seasons. CONSTITUTION:(1) Calluses or an adventitious embryos are induced. A medium for example, Murashige-Skoog medium added with 10-50 g/l of sucrose, is used. (2) The calluses are reproduced in a liquid medium for reproduction, at 25 degree C and under an illumination by a fluorescent light. (3) The derived calluses are transplanted into a medium for redifferentiation. The medium for redifferentiation is for example, a solid medium from the Murashige-Skoog medium added with 2% of sucrose together with hormone such as auxin and the like, and the process is performed at 15-35 degree C, under a illumination of 1000-10000lux. The objective seedlings are produced by the above three processes.</p> |
