PURPOSE:To obtain a hematopoiesis regulation factor useful as a medicine for improved blood cell vital abnormality follouring various blood cell diseases and use of anti-cancer medicines, having promoting action on multiplication of premature cells such as hemateopoietic stem cell, produced by strama cell and fibroblast. CONSTITUTION:A stroma cell, a fibroblast, etc., is cultured in a medium containing 10% bovine serum. At a point of time when the medium is made confluent, the medium is replaced. On the third day, a supernatant liquid of the culture is collected, cultures in a medium containing lipopolysaccharide (LPS).TPA and stimulated. A supernatant liquid of the culture is concentrated by affinity chromatography and gel filtration and purified to give the objective hematopoises regulation factor which is a protein adsorbed on silica gel, DEAE sephacel and heparin agarose in a dissolved state in 10mM phosphate buffer solution at pH 7.0, has about 80,000 molecular weight, supports multiplication of interleukin-3 (IL-3)-dependent cell strains but its multiplication supporting ability is not influenced by an anti-IL-3 antibody. |
