PURPOSE:To obtain a new enzyme useful for producing penta-N- acetylchitopentaose suitable as a substrate for measuring lysozyme activity in a clinical examination from colloidal chitin selectively and in high yield. CONSTITUTION:Chitinase A. Action: acting on colloidal chitin to form N- acetylglucosamine and N-acetylchitooligosaccharide having 2, 3 and 5 degree of polymerization. Optimum pH: pH 5 and pH 9 (substrate is colloidal chitin) at 30 deg.C. Optimum temperature: pH9 45 deg.C at (substrate is colloidal chitin). pH Stability. stable pH5-10 at 30 deg.C for 120 minutes. Thermal stability: stable by treatment at pH9 at 40 deg.C for 1 hour. Molecular weight: 66,000 (SDS-PAGE method). Isoelectric point: 4.3. The enzyme is obtained by culturing a fungus belonging to the genus Vibrio capable of producing the chitinase A [e.g. Vibrio alginolyticus TK-24 (FERM P-12,011)] and isolating the enzyme from the culture mixture. |
