PURPOSE:To provide a new DNA useful for production of mink growth hormone. CONSTITUTION:This invention is a recombinant DNA in which a DNA coding an amino acid sequence of the formula is inserted into a vector DNA (e.g. plasmid, pUC119DNA). From an RNA prepared from mink-derived hypohysis, a cDNA is synthesized and the resultant cDNA is integrated into a vector DNA to obtain various kinds of transformants and to obtain a 700bp growth hormone cDNA from a cDNA gene bank library. The resultant cDNA is treated with restriction enzymes EaeI and BamHI and the obtained 650bp CDNA fragment (a growth hormone gene having a structure in which base sequences coding two amino acids from the N-terminal of the mature growth hormone are removed) is used in combination with n promoter, a vector DNA and a synthetic DNA having a base sequence coding two amino acids from the N-terminal of mink growth hormone gene, the presequence and seven prosequences of a neutral protease II and Kex 2 recognition amino acid sequence, thus affording the objective recombinant DNA. |
