PURPOSE:To obtain a new glutamic acid dehydrogenase excellent in the heat resistance and salt resistance and useful for clinical diagnosis, etc., by culturing a glutamic acid dehydrogenase-producing bacterium belonging to the genus Bacillus and subsequently collecting the enzyme. CONSTITUTION:A glutamic acid dehydrogenase-producing bacterium belonging to the genus Bacillus, such as Bacillus.acidocaldarius is cultured, and the produced enzyme is collected. The objective dehydrogenase has following characteristics; (i) action: catalyzes the oxidative amino-removing reaction of L-glutamic acid in the presence of nicotinamide adenine dinucleotide; (ii) substrate specificity; exhibits a substrate specificity to L-glutamic acid; (iii) optimal pH: 6.5-8.5; (iv) stable pH range: 5.0-13.0; (v) the range of suitable action temperatures: up to 75 deg.C; (vi) thermal stability (inactivation condition): stable up to 70 deg.C, and stable up to 95 deg.C when sodium chloride is present in an amount of 2M; (vii) inhibitor: inhibited with 1 mM mercuric chloride; (viii) mol.wt.: approximately 310000 by a gel filtration method. |
