<p>PURPOSE:To produce a cardiac glycoside such as digitoxin in high productivity by adding 5beta-pregnane-3,20-dione to a culture medium of a culture strain of foliar organ of a plant to produce a cardiac glycoside, carrying out tissue culture and collecting a product from the culture mixture. CONSTITUTION:Leaf pieces of a plant (e.g. Kedigitalis) of the genus Digitalis to produce a cardiac glycoside such as digitoxin or digoxin are placed on an agar medium containing indoleacetic acid and benzylaminopurine, subjected to stationary culture at 25 deg.C at a light place having 1,500-3,000 lux to form calluses, a differentiated foliar organ is taken out from part of the calluses. The organ is repeatedly subjected to subculture at every 3 to 4 weeks to give a culture strain of foliar organ. Then the culture strain is subjected to shaking culture in a liquid medium for tissue culture containing 5beta-pregnane-3,20-dione and/or 5beta-pregnane-3beta-ol-20-one at 25 deg.C at a light place for 9 days, the culture mixture is filtered, the filtrate is freeze-dried and the dried substance is extracted with ethanol to give the cardiac glycoside.</p> |
