Prodn. of probes specific for human chromosome 21 includes the step of amplifying the genomic DNA sequences of a hybrid human-rodent cell line contg. chromosome 21 sequences as the only human sequences. Amplification is effected using the primers Alu TC65 and Alu 517 (Proc. Natl. Acad. Sci. USA, 86,6686, 1989). USE - The probes are useful for diagnosis of trisomy 21 (Down's syndrome) by determining the number of copies of chromosome 21 in human cells, esp. metaphase cells or interphase cell nuclei.
