摘要 E.coli expression vector contains (i) part of the DNA of the Thermus (T.) thermophilus (therm.) 26.8 kd-NADH-oxidase (nox) gene having an amino acid (AA) sequence defined in the specification, (ii) a DNA sequence hybridising with (i), pref. under 6xSSC, 1h wash with 0.6xSSC, 0.1% SDS, more pref. with 4xSSC and 1 h wash with 0.1% SDS, 0.1% SSC, each at 62-66 deg. C, and (iii) the AA sequence with either 10 AA deleted, added or substd.. Also claimed is the use of an expression vector or of E. coli for the prodn. of nox or an exzyme having that activity. USE/ADVANTAGE - Nox is used as a biosensor. The enzymes derived from thermophilic organisms are very stable and owing to a longer life span they are preferred in tests.
