<p>PURPOSE:To provide a novel monoclonal antibody for e.g. diagnosing Alzheimer' s disease, common to senile macula amyloid precursor protein APP 770, 751 and 695 and capable of specifically recognizing part of the section other than the sequence from the membrane penetration region to C-terminal. CONSTITUTION:Human senile macula amyloid precursor protein is boiled for 3 min in a surfactant-contg. phosphoric acid buffer solution followed by acetone treatment, and the resulting protein, in combination with an adjuvant, is administered to Balb/c mice through subcutaneous injection to immunize the mice; after the final immunization, the mice's splenocytes are collected and fused with myelolipoma strain cells followed by culture in a HAT medium, thus obtaining a hybridoma. Thence, the hybridoma is screened to select a clone capable of producing anti-human senile macula amyloid precursor protein antibody, the clone being then cloned by e.g. the limiting dilution analysis to effect monocloning. The resulting hybridoma is then incubated, thus obtaining the objective novel monoclonal antibody.</p> |
